<原著>脳卒中易発症性高血圧自然発症ラットの末梢血行動態に与えるカルシウム拮抗薬の影響

The hemodynamic effects of calcium antagonists have been studied on conscious 6-month-old stroke-prone spontaneously hypertensive rat (SHRSP) and on normothesive Wistar Kyoto rat (WKY). The blood pressure and heart rate were measured before and after administration of calcium antagonists NB-818 and nifedipine respectively. Rats were grouped as follows : 10 nmoles/kg i.v. (Group 1) and 30 nmloes/kg i.v. (Group 2) of NB-818 in SHRSP ; 30 nmloes/kg i.v. (Group 3) of nifedipine in SHRSP and 30 nmloes/kg i.v. (Group 4) of NB-818 in WKY. Cardiac index (CI) and regional blood flow were also measured by the radioactive microsphere method. Mean arterial pressure was significantly lowered by both agents. Compared with the vehicle group, CI was increased in the NB-818 treated SHRSP, concomitant with significant decrease in total peripheral resistance index (TPRI). The cerebral vascular resistance was lowered by 41% in Group 1,and 47% in Group 2,with corresponding increases in cerebral blood flow by 46% and 32%, respectively. However, the cerebral flow fraction of the cardiac output (% CO) was not increased. In Group 3,the cerebral vascular resistance was lowered by 25%, cerebral blood flow was increased by 10%, and cerebral flow fraction of cardiac output was decreased by 20% compared with the vehicle groups. In Group 4,cerebral vascular resistance was decreased by 53% and cerebral blood flow was increased by 79%. In the kidney, the peripheral resistance was decreased by 36% in Group 1,and 48% in Group 2,respectively. In Group 3,vascular resistance was decreased by 30% and regional blood flow was increased by 28%. In gastrointestinal tract (GIT), regional blood flows were increased by 65% in Group 1,and 79% in Group 2. In WKY, regional blood flow was increased in the kidney and the GIT compared with the non-treated group as well as in SHRSP. In conclusion, NB-818 is a useful agent for treatment of cerebrovascular diseases such as hypertension, in which regional blood flow in the brain and in the major visceral organs are eventually decreased. This agent can reduce vascular resistance even in severe hypertensive SHRSP as well as in normotensive rats.
