We report the stoichiometry between the number of single strand breaks induced by a rat liver endonuclease and the number of thymine dimers and 2-acetylaminofluorene (2-AAF) adducts released. We demonstrate that the same proteins act on both UV irradiated and 2-AAF bound DNA. We exclude the possibility that the enzyme acts as a glycosylase. We provide evidence that the enzyme yields a 3'OH and 5'PO_4 at the site of the single strand break. We establish that the enzyme has a broad specificity and also acts on X-irradiated and methylated DNA.
