上皮増殖因子 (EGF) ドメインはユニークなO-結合型糖鎖による翻訳後修飾を受ける。EGFドメイン上のO-型糖鎖として知られるO-フコースやO-グルコースは、多くの血漿糖タンパク質やシグナリング分子に見出されており、とりわけNotchシグナリング経路における生物学的機能がこれまでに示されてきた。我々は最近、EGFドメインにおける新規の糖鎖修飾としてO-GlcNAcを発見した。そしてこの修飾が、ショウジョウバエNotch受容体のEGFドメインにおける5番目と6番目のシステイン残基の間にある、保存されたSer/Thr残基に起こることを見出した。この細胞外のO-GlcNAc修飾に関わる責任遺伝子を単離し、その機能を探ることで、分泌タンパク質や膜タンパク質、そして細胞外環境におけるO-GlcNAc修飾の生物学的役割が明らかになるであろう。
