We studied the influence of the substrate topography substrate on the cell respondence. Topographical cues, independent of biochemistry, generated by the polydimethylsiloxanes (PDMS) substrate may have significant effects on cellular behavior. Mouse bone marrow stromal fibroblasts were examined on grooved substrata of equal groove and ridge width (100 μm, 30 μm depth). The phase contrast light microscopy revealed the attachment and orientation of cells on the materials used and indicated that the fibroblasts prefer to grow to confluent on the top of the ridges. This showed that the topographical surface had a profound effect on cell alignment and migration.
