Interrelationship between Lipogenesis and Creatine Biosynthesis

Countenance of young myopathics, whose subcutaneous fat tissue overlies atrophic muscles abundantly, shows facies myopathica. In these individuals, muscle components are decreasing, whereas lipogenesis may increase.<BR>The present studies are undertaken in an attempt to elucidate whether lipogenesis counteract or not biosynthesis of creatine, which is one of the muscle components.<BR>Wistar strain albino rats weighing 80g are used. By adding creatine in the diet, kidney glycine amidinotransferase, which reacts on the first step in creatine biosynthesis, is inhibited by creatine as the end product. In the same creatine-fed individuals as described above, incorporation of <SUB>14</SUB>C-acetate intraperitoneally administered into liver lipid fraction exhibits an increase in the distribution patterns similar to that of a creatine-free basal diet. The increase of <SUB>14</SUB>C-acetate incorporation by adding creatine is not observed when they are fed on 10% fat diet, but this increase is exhibited on fat-free diet. The creatine effect may be associated with lipogenesis.<BR>Therefore, liver glucose-6-phosphate dehydrogenase and acetyl CoA carboxylase, involved in lipogenesis, are estimated. The result is that both enzymes show high activities on fat-free and sucrose-rich diet, but much higher ones on the creatineadded sucrose-rich diet. Those effects of creatine on lipogenic enzymes, specially on acetyl CoA carboxylase, appear with or without choline in the diet. So, creatine may accelerate lipogenesis directly, or partly due to the increased larger pool of choline in accordance with diminished transmethylation on the second step in creatine biosynthesis as a result of feedback inhibition by creatine. These findings lead us to the suggestion that an individual with creatine in excess?myopathics who are associated persistently with creatinuria is predisposed to lipogenesis.
