Four species of trehalase have been isolated from rabbit kidney cortex. These four species are similar in the enzymatic activity, but different in molecular weight, carbohydrate content, and susceptibility to HgCl<SUB>2</SUB>.<BR>It is proposed from some evidences that physiological function of renal trehalase is a glucose transporter (glucose carrier protein) in the brush border membranes of proximal tubules. First, the substrate-binding site of renal trehalase contains two subsites; one is phlorizin-binding site, the other is phloretin-binding site. These findings are similar to the results of Alvarado (1967); namely, a phlorizin-binding protein associated with glucose transport mechanisms has two binding sites in the active sites; phenyl site and glucosyl site Secondly, membrane topography of trehalase is similar to the glucose transporter, which is a trans membrane protein. Thirdly, renal trehalase is activated by Na<SUP>+</SUP> which activate glucose transport, and inhibited competitively by glucose.
