In 1963, soil samples were collected from eight plots in four areas of Aomori prefecture and from seven plots in three areas of Hokkaido. These plots were selected to include both rough-bark orchards and rough-bark free orchards. The soil profiles were investigated in each plot, and the soil samples were analysed for their chemical and physical properties. Also the mineral constituents of the apple leaves and shoots in each plot were analysed, and the following points could have been elucidated as to the physiological mechanism of the rough-bark symptom.1. The rough-bark symptoms occurred in high frequencies where the soil reduction was enhanced due to the strong acidity or ill drainage or both.2. As compared with rough-bark free orchards, the rough-bark orchards contained more manganese in high concentration in the surface soil. The concentration of exchangeable Mn (including water soluble Mn) and of easily reducible MnO2 were 15.9 and 9.9ppm; 266 and 139ppm, respectively, in the soil of the 1st and 2nd layers in rough-bark orchards. Furthermore, it was surmised that manganese was more closely associated with the disorder in its easily reducible state than in its exchangeable state.3. The results of analyses in leaves and shoots showed that there was no difference in the contents of phosphorus, potassium, calcium, magnesium and iron between the healthy and unhealthy trees of the same variety. Only in the manganese content they differ ed conspicuously: the rough-bark trees contained much higher amounts of this mineral than did the healthy ones. This is in agreement with the results of our previous report (AOKI and OKUSE, 1964). On the other hand, the effect of stock on the incidence of the disorder was less clear in fully-grown trees than in young ones.4. When the manganese contents of each variety grafted on the same kind of stock were compared among healthy and diseased orchards, it became apparent that the soil conditions drastically influenced the quantity of the mineral absorbed in the plant body. Namely, the contents of manganese of leaves and shoots in the rough-bark orchards were three to four times as much as those in symptom free orchards. There were no varietal differences in manganese absorption and in the outbreaks of the symptoms.
