DISC ELECTROPHORETIC ANALYSIS OF GROWTH HORMONE AND PROLACTIN IN THE ANTERIOR PITUITARY OF MOUSE

Present experiment was carried out in order to determine growth hormone and prolactin in the anterior pituitaries of mice by disc electrophoresis at pH 9.5. The slowest migrating and intermediate bands were identified as growth hormone and albumin respectively. A rather faint band just in front of the albumin band was considered to be a prolactin band. It was demonstrated that the linear relationship existed between the dose of bovine growth hormone (NIH-GH-B<SUB>12</SUB>) and the optical density of the band obtained by electrophoresis. Electrophoresis at pH 4 was also carried out but with no better result. Disc electrophoresis is considered to be a very convenient method for at least semiquantitative analysis of growth hormone in the anterior pituitaries of mice, one gland being generally sufficient for satisfactorily clear detection, and very sharp band is obtained. On the other hand, this method is not always suitable for analysis of the anterior pituitary prolactin in this species, and some other devices would be necessary.
