Some immunological properties of rabbit antiserum to mouse prolactin (MPL) were examined as a fundamental experiment for the study on the role of prolactin in mouse mammary gland. Antiserum to MPL was obtained by immunizing a New Zealand White female rabbit with MPL preparation. Precipitin lines developed when unabsorbed antiserum was reacted with normal mouse serum (NMS) and the extracts of pituitary, liver, kidney, spleen and brain by the method of Ouchterlony agar gel double diffusion. The antiserum absorbed with NMS and spleen extract (Absorbed Antiserum) showed the positive reaction only with the pituitary extract. Absorbed Antiserum reacted with MPL preparation and a single precipitin line developed between them by agar gel double diffusion. A single precipitin arc was observed at the corresponding site of MPL band in the immunoelectrophoresis of Absorbed Antiserum using polyacrylamide gel after electrophoresis of the pituitary homogenate. Furthermore, the antiserum inhibited markedly the biological activity of MPL. All the results have demonstrated that Absorbed Antiserum obtained has a single antibody specific to MPL. Absorbed Antiserum did not cross-react with bovine prolactin or the pituitary extracts of caprine and rabbit, but cross-reacted with rat pituitary extract. Antigenic identity between prolactin from mouse source and that from rat source was incomplete, because the precipitin line formed a spur.
