Effect of trifluoperazine on phagocytosis of cultured Kupffer cells.

The role of the Ca<SUP>2+</SUP>-calmodulin system was studied on the phagocytosis of cultured rat Kupffer cells isolated by collagenase digestion and Percoll density gradient centrifugation using calmodulin inhibitor; trifluoperazine.<BR>The endocytosis of polystyrene beads into cultured Kupffer cells was inhibited by the preincubation with trifluoperazine as a dose dependent manner and this inhibitory effect was reversible when changed culture medium without trifluoperazine. Morphologically, pseudopodia on cell surface and actin filaments accumulation beneath pseudopodia were disappeared by preincubation of trifluoperazine.<BR>These results suggested that the actin-myosin system through the Ca2+-calmodulin system played an important role on the phagocytotic movement of Kupffer cells as similar to the contraction system of other non-muscle cells.
