Analysis of DNA content and s-phase fraction in colorectal cancers.

A retrospective study has been made to determine whether DNA ploidy pattern can be a factor to reflect progress and prognosis of colorectal cancer 122 patients undergone resection. DNA ploidy pattern was obtained by flow cytometry by use of paraffin-enbedded pathological materials. As a result, it was found that 5 year survival rates in the cases under gone overall or curative resection among aneuploid cases were significantly low (p <0.05), compared with diploid cases. A trend liable to cause lymphatic or hepatic metastasis was also observed. <BR>Next, an assessment was made regarding the cause of difference in malignancy between aneuploid and diploid using cell kinetic technique. The resected organs were put to ex-vivo organ perfusion to have cancer cells take in bromodeoxy-uridine, and S-phase cell fraction was calcurated by flow cytometry. As a result, S-phase cell fraction was found as 19.3% in diploid and 20.8% in aneuploid, showing no significant difference between the two.
