The apical portion of the cauliflower inflorescence has a high DNA content. Soluble chromatin can readly be isolated, showing that the apical portion is a suitable source for the mass preparation of isolated chromatin. Two methods for the determination of the histone content of isolated chromatin were used. RNA synthesizing capacities were also examined. The degree of template activity does not depend on amount of histone estimated in the TCA-precipitate, but does depend on the amount of histone determined in chromatin in the soluble state. Condensed- and extended-chromatins were separated and extended chromatin had four times as high template activity as did condensed chromatin.
