Efficacy of the Extracorporeal Granulotrap G-1 Column in a Rabbit Model of Induced Arthritis(Workshop1-12)

A rabbit model of induced arthritis was prepared to assess the efficacy of an extracorporeal type column packed with cellulose acetate beads (G-1 column). Rabbits were sensitized by intradermal injection of 4mg ovalbumin (OVA) on two occasions, two weeks apart followed by an intra-articular injection into one knee joint of five mg OVA, 5 days later (i.a OVA). Arthritis was developed one day after i.a OVA as evidenced by marked joint swelling and the extensive infiltration of leucocytes (>95% granulocytes and monocytes) into the knee joint. Ten minutes after i.a OVA, the rabbit blood was passed through an 18 ml capacity G-1 column at 2 ml/min for 1 hr. About 32.4% of the leucocytes in the perfused blood were adsorbed onto the G-1 column beads, 80% were granulocytes and monocytes. Observation revealed suppression of joint swelling (p<0.001), leucocyte infiltration into the joint (p<0.001) and proteoglycan loss from the joint cartilage (p<0.05). It is suggested that the efficacy of this treatment is attributable to the removal or otherwise suppression of hyperactive inflammatory leucocytes by the G-1 column.
