Biological Fate of Orally Administered Naringin and Naringenin in Rats

The fate of orally administered Naringin (NI) and Naringenin (NE) was studied in rats. Structural investigations of the metabolites excreted in bile and urine, following NT and NE administration, revealed that both drugs were transformed to the same metaholites. The major biliary metabolites were NE 7-glucuronide, NE 7-sulfate 4'-glucuronide and NE 7-glucuronide 4'-sulfate. On the other hand, the major urinary metabolites were NE 4'-glucuronide and NE 7,4'-disulfate, in addition to the foregoing NE 7-glucuronide, all of which were found in plasma as the major metabolites. Studies on the plasma concentrations and urinary and biliary excretion of the metabolites following drug administration demonstrated that NE was more readily absorbed than NI, giving rise to higher concentrations of the metabolites in plasma and urine as well as in bile. These findings indicated that NI was absorbed as NE after hydrolysis.
