Wood part of Agarwood is characterized by that it includes phloems differentiated in xylem tissues, in addition to the usual secondary phloems derived from cambium. In lower grade Agarwood samples resinous cells were identified only in the phloem tissues in xylem when examined under a reflection fluorescence microscope, while in higher grade samples resinoid, containing phenethylchromones, was observed extending from phloem into medullary rays and then into xylem tissues, as the resinoid content increased. The colour slides of the resinous tissues taken with a fluorescence microscope were subjected to spectrophotometric analysis to produce visuable absorption curves. The resinoid in phloem and those in xylem were shown to give the same thin layer chromatogram and densitometry pattern. Acetone extracts from several resinoid-containing tissues of Aquilaria malaccensis Lam. were found to contain no phenethylchromones and to be different in composition from that of Agarwood by TLC and densitometry.
