The Stimulatory Effects of Oriental Crude Medicines on Interleukin-2 Gene Expression in a Human T Cell Line

• Oriental crude drugs were tested for enhancement of interleukin-2 (IL-2) gene expression in Jurkat cells, a human T cell line. The aim of this screening was to identify small chemicals that increase IL-2 expression that can be used as therapeutic reagents in immune therapy, such as tumor adoptive therapy, and that would also act as chemical probes in the pharmacological investigation of the intracellular signaling mechanisms directing IL-2 gene expression. We determined the change in IL-2 mRNA level in sample-treated cells by competitive-PCR-based quantitative PCR. Of 102 crude drugs, 16 drugs increased IL-2 mRNA expression more than 2-fold when compared with the vehicle-treated cells. In particular, Rhei Rhizoma, Nupharis Rhizoma, Caryophylli Flos and Magnolia Cortex enhanced IL-2 gene expression more than 10-fold.
