It is well known that γ-oryzanol includes ferulic acid esters such as cycloartenyl ferulate, 24-methylenecycloartanyl ferulate, stigmasteryl ferulate, campesteryl ferulate and (β-sitosteryl ferulate. Since ferulic acid esters from Notopterygii Rhizome (Notopterygium incisum) and Atractylodis Lanceae Rhizome (Atractylodes lancea) inhibited the activities of lipoxygenase (LPO) and cyclooxygenase (COX), we examined the effects of ferulic acid esters from γ-oryzanol on LPO and COX activities in this study. All ferulic acid esters used in this study inhibited the activity of LPO (IC_<50>: 15-34 μM). Moreover, cycloartenyl ferulate and stigmasteryl ferulate also inhibited the activities of both COX-1 and COX-2. Then, to evaluate the anti-inflammatory activity of γ-oryzanol in vivo, we examined the effects of γ-oryzanol on adjuvant-induced arthritis in rats. It was revealed that γ-oryzanol inhibited the increase in swelling of the hind paw in a dose dependent manner (1-100 mg/kg). These results suggested that γ-oryzanol had therapeutic potential for inflammatory disease.
