肉〓蓉の生薬学的研究(第2報) : Cistanche属植物の成分の比較

In our previous paper, we reported that Cistanche plants from Asian countries and commercial samples of the crude drug Roucongrong (肉〓蓉) from China could be classified into three types by the morphological and anatomical characteristics, i.e., C. deserticola (Cd) type, C. tubulosa (Ct) type and C. salsa (Cs) type. In this paper, we investigated the chemical components in Cistanche plants from Asia to find out if they could be used for the identification of the botanical source of the Chinese crude drug "Roucongrong" on the market. Seven phenylethanoid glycosides (PeG) in the specimens were quantitatively determined by HPLC. The results were as follows: (I) Samples belonging to the Cd type and Cs type contained cistanosides A and C, and a trace of tubuloside A, whereas those belonging to the Ct type contained a quantity of tubuloside A but not cistanosides A and C. Thus, the results of chemical analysis did not controled the morphological classification of the previous report. (II) The total amounts of PeG in the Ct and Cs type plants were larger than those in the Cd type plants. The total PeG content in the crude drug samples from Cd type plants in the Chinese market was very small. (III) The total PeG content in the callus derived from C. deserticola and C. tubulosa seeds were about 50 fold and 3 fold that of the mother plant, respectively. However, the PeG content in the callus of C. deserticola decreased significantly when dried slowly at room temperature for 4 weeks. The result of this experiment suggested that the considerably lower PeG content of the crude drug, compared with that of the original material, might be caused by the preparation procedures.
