Exercise induces eustress both physically and mentally and seems to contribute to the extension of healthy life and quality of life (QoL). However, it remains uncertain how exercise modulates stress responsiveness in elderly adults. Recently, the measurement of salivary alpha-amylase (sAA) has been considered to be a useful tool for evaluating stress responses. We investigated the association between physical activity and psychophysiological stress in elderly adults using a portable hand-held monitor to investigate sAA. sAA levels were measured in 20 healthy volunteers before and after walking. Self-reported affective responses were also assessed using the Profile of Mood States (POMS)-brief Japanese version before and after walking. The mean value of the sAA activity was significantly higher in the elderly adults (n=10, 99.05±21.1kU/l) than in the young adults (n=10, 34.4±5.9kU/l) (p<0.05). However, no significant difference was found between before and after walking for sAA levels. All negative feeling scores significantly decreased after walking compared with before walking for young adults. 'Vigor' scores significantly increased after walking compared to those before walking only in the young adults (p<0.001). 'Confusion' scores improved after walking for elderly adults. sAA activity after walking for elderly adults correlated positively with 'tension-anxiety' (r=0.667, p<0.05), 'depression-dejection' (r=0.789, p<0.01), and 'fatigue' (r=0.827, p<0.01), as determined by the POMS-brief. These results suggest that moderate aerobic exercise such as walking has psychological beneficial effects and leads to healthy brain aging. sAA activity may be a useful, non-invasive index to evaluate psychophysiological stress before and after exercise in elderly adults.
