EFFECTS OF INTRACEREBROVENTRICULAR MORPHINE AND ENKEPHALINS ON THE URINE OUTFLOW IN ETHANOL-ANAESTHETIZED RATS

Effects of intracerebroventricularly (i.c.v.) administered morphine and met- and leu-enkephalins on urine outflow were investigated in i.c.v. perfused rats. Morphine, when injected i.c.v. at the dose of 1 nmole, produced naloxone-reversible antidiuresis. Met-enk and leu-enk, when injected i.c.v. at the dose of 0.1 and 1 nmoles increased urine outflow. The effect of met-enk was not dose-dependent but inhibited by naloxone, suggesting that diuresis may be mediated through opiate receptor sites in the central nervous system. Since the diuretic effect of met-enk was diminished by propranolol and reserpine, it was suggested that met-enk released small amounts of noradrenaline into synaptic cleft to activate β-adrenoceptors. Met-enk did not change sensitivity of α-adrenoceptors, since an antidiuretic effect of noradrenaline was not altered by met-enk. Similarly, met-enk did not alter antidiuretic effects of histamine, GABA and glutamate, suggesting that met-enk increased urine outflow not through inactivation of receptor sites for these agents.
