Effects of prostaglandins on central regulation of water metabolism were discussed. The hypothalamus where PGs are present is important region for regulation of water metabolism. The effectiveness of PGE_2 was dependent upon the region where it was a applied. PGE_2 induced diuresis which was mediated by β-adrenoceptors in the ventromedial nuclei. On the other hand, the antidiuretic effect of PGE_2 which might be due to promoted secretion of antidiuretic hormone was mediated by α-adrenoceptors in the supraoptic and paraventricular nuclei. Of interest is that unlike PGE_2, PGH_2 which is a precursor of PGE_2 possessed the antidiuretic effect but not the diuretic effect. Since PGD_2, PGI_2 and TXB_2 were without effect, an importance of primary PGs in this field can be stressed.
