We have instituted studies of endogenous digitalis-like substances (EDLSs) in the organs and tissues of Bufo marinus. Thirteen compounds were isolated by reverse-phased HPLC, and the structures were determined by NMR and mass spectrometry.Polyhydroxylated bufadienolides were isolated from the skin and the eggs, glucuronyl bufadienolides from the bile, and conjugated bufadienolides from the serum. In addition, two novel compounds named marinosin and marinoic acid were identified in the skin. Marinosin differed from typical bufadienolides in having its B ring in a boat, rather than chair, form. Marinoic acid differed from typical bufadienolides in that its C and D rings. The inhibition of Na^+,K^+-ATPase enzymatic activity, and of [^3H]ouabain binding activity were tested. All of these compounds showed similar activities; genins were the most potent inhibitors; conjugated bufadienolides were potent; marinosin and marinoic acid were less potent; and glucuronyl bufadienolides were very weak inhibitors. These EDLSs, especially marinosin and marinoic acid, may be important in the identification of a physiological role of bufadienolides in the toad, Bufo marinus, as well as in the identification of EDLS in mammalian species.
