The associations of the specific HLA-DRB, HLA-DQA, and HLA-DQB alleles and haplotypes with type 1 diabetes in Japanese children

Objectives : In this study, we investigated the association of human leukocyte antigen (HLA) class II genes and type 1 diabetes in 30 unrelated patients with type 1 diabetes and 97 healthy controls from a Japanese origin using case-control study. Materials and Methods : DRB1-DQA1-DQB1 alleles and haplotypes were determined by the PCR-sequencing-based typing (SBT) method reported by Nomoto et al. Results : The phenotypic frequencies of DRB1*0901-DQA1*0301-DQB1*0303 haplotype were significantly increased in patients with type 1 diabetes. Furthermore, we also found the homozygotes DRB1*0901/*0901 genotype was the highest relative risk for type 1 diabetes and the phenotype frequency of DRB1*0901/*0405 genotype was also increased significantly in type 1 diabetes. Protective haplotypes from type 1 diabetes revealed some peculiarities in Japanese children : the phenotypic frequencies of DRB1*0101-DQA1*0101-DQB1*0503 and DRB1*15021-DQA1*0103-DQB1*0601 haplotypes were significantly less common in patients with type 1 diabetes, which gave dominant protection against type 1 diabetes. Conclusion : Our observations confirmed the association of specific HLA-DQ and HLA-DR regions with type 1 diabetes, and might underline several characteristic genetic backgrounds that distinguish Japanese juvenile patients from Caucasians.
