Loss of heterozygosity at a cellular senescence locus on human chromosome 2 in various types of tumors

Since the cellular senescence trait is thought to be resulted from mutation, deletion, or inactivation of a gene or genes that requires for the cellular senescence program in normal cells, the senescence program could be functionally restored by the transfer of a specific normal human chromosome. Thus, the microcell-mediated chromosome transfer (MMCT) has mapped chromosomal regions that contain a putative cellular senescence gene(s) to some 10 chromosomes. A normal human chromosome 2 restored a mortality and induced cellular senescence in the human cervical carcinoma cell line SiHa, and the candidate region for the cellular senescence gene has previously been mapped to the 2q37 region via MMCT. Therefore, the cellular senescence gene(s) is expected to have been lost in various tumor tissues like other tumor suppressor genes previously identified. Here, we examined the frequency of loss of heterozygosity (LOH) at the 2q37 (D2S172) and the other unrelated region 2p13-16 (D2S134). We analyzed genomic DNAs from 7 different types of tumor tissues in 162 patients, and the LOHs at D2S172 and D2S134 were detected in all of the tumor types and in 2 of the 7 tumor types examined, respectively, which is consistent with the mapping information for the cellular senescence gene by the previous MMCT experiment.
