The effect of methanol and KCl, NaCl, MgCl_2, CaCl_2 or SrCl_2 on the frequency of respiration-deficient (RD) mutation in yeast, Saccharomyces cerevisiae, induced by MnCl_2 was investigated. The frequency of RD mutation induced by 1×10^<-2>M MnCl_2 was increased from 9.5% to 59.6% by the addition of 4% methanol to the culture medium. However, the frequency of RD mutation induced by MnCl_2 was decreased by the addition of 1×10^<-2>M MgCl_2, CaCl_2 or SrCl_2 in the presence and- absence of methanol. The growth-inhibitory effect of MnCl_2 was also partially reversed by the addition of these divalent metal compounds, whereas it was enhanced by methanol. Monovalent metal compounds, KCl and NaCl, had little effect on the RD mutation or growth inhibition by MnCl_2. Uptake of Mn^<2+> by yeast cells was increased significantly by the addition of 4% methanol, and was decreased by MgCl_2, CaCl_2 or SrCl_2. Among these compounds, CaCl_2 decreased the uptake of Mn^<2+> most effectively.
