Complement Activating Bactericidal Factor, Ra-Reactive Factor, of Rats : Similarity to That of Mice

Ra-reactive factor (RaRF) is a complement-dependent bactericidal factor that binds specifically to Ra chemotype strains of Salmonella. Here we investigated certain characteristics of RaRF of rats. The rat RaRF activated complement in a dose-dependent manner. C1 depleted serum was as effective as the undepleted serum in the hemolysis of Ra lipopolysaccharide-coated erythrocytes (ELPS) which were sensitized with RaRF, indicating that C1 is not responsible for the complement activation induced by the factor. On the other hand, the RaRF bound to ELPS activated C4 and C2, forming C3 convertase on the cells. There was a diisopropylphosphorofluorate-sensitive C4 activating factor in the RaRF complex. It was demonstrated by density-gradient centrifugation and polyacrylamide gel electrophoreses that the rat RaRF complex contained a serine proteinase together with two major polypeptides corresponding to P28a and P28b of mouse RaRF. These results indicate that rat RaRF resembles mouse RaRF not only in its function but also in structure.
