Light-and Electron Microscopic Observations of a Strain of Free-living Nematodes, Rhabditidae tokai

The morphological structure of a strain of free-living nematodes, Rhabditidae tokai, was observed under light-and electron microscopes. Through the transparent cuticle layer, the following organs could be observed under a phase-contrast microscope: a digestive tract consisting of an esophagus with two bulbs and an intestine with a large number of granules; an ovary and an uterus in females holding premature egg cells which were growing towards the vulva and a testis in males with a large number of sperm cells and a copulatory spicule. Photographs taken by a scanning electron microscope revealed the shape of the head portion and a regular structure of cuticle layer with lateral pleats. Fine structures of the pharynx and body portions could be observed under a transmission electron microscope. They were very similar to those of other nematodes including Caenorhabditis elegans.
