Light-and Electron-Microscopic Study on the Localization of Human Chorionic Gonadotropin in Chorionic Villi and Placenta by an Enzyme-Labeled Antibody Technique

The localization of H. C. G. in the villi and the placenta, and its significance on continuance of pregnancy were investigated. Twelve villi and placentas from 5-weeks to fullterm pregnancies and an uterus of 9-weeks pregnancy, which was extirpated because it was affected with myoma uteri, were used. The enzyme-labeled antibody method and the various blocking tests were performed and eliminated contaminations such as nonspecific reactions, which were concomitant in prior methods. The following results were obtained: (1) By an area-wise study of the villi, prominent reaction products were found in the junction of the villi and decidua, syncytial sprout, syncytial cell on the decidua and the syncytial cell in the decidua. (2) Electron microscopy located reaction products in the syncytial cell membranes and cisterna of the R. E. R. and the outer nuclear membrane. (3) The marked appearance of reaction products in the growing part of the villi and in the syncytial cells in osculation with the maternal body leads of the interpretation that H. C. G. acts on the maternal body directly and indirectly for the villi to grow and display their functions. (4) According to the length of pregnancy, strong reactions were noticed at 5 to 12 weeks pregnancy, but the reaction after 20 weeks of pregnancy was attenuated and irregular. The full-term placentas gave only an extremely feeble reaction.
