Endostatin, an endogenous angiogenesis inhibitor derived from type XVIII collagen, is highly promising as part of cancer therapy. Endostatin shows a multi-binding ability to tropomyosin, heparin, perlecan, glypican. integrin, zinc, laminin, and fibulin. Among these, integrin and glypican are considered to be functional receptors of endostatin, although their binding affinities arc low to moderate. The enzymatic activity of matrix metalloproteinases is inhibited by endostatin, which binds to their catalytic domains. The anti-tumor activity of endostatin is mediated not only by its anti-angiogenic activity, but also manifests itself in its direct effect on tumor cells. Despite the many studies published on endostatin, its mechanisms seem to be complicated and still remain to be elucidated. The possibility of substituting synthetic peptides for endostatin is also introduced and discussed.
