Carboxylation and Detoxification of Xylulose Bisphosphate by Spinach Ribulose Bisphosphate Carboxylase/Oxygenase

The time-dependent, slow inhibition of ribulose bisphosphate carboxylase (RuBisCO) in the absence of ribulose bisphosphate was dependent on the concentrations of RuBisCO and xylulose bisphosphate (XuBP). When incubated with excess XuBP, RuBisCO lost its activity gradually with incubation time. When RuBisCO of the concentration of 1.5μM was incubated with 20μM XuBP, the activity was inhibited for the initial 10 minutes, after which the activity recovered gradually with time. The recovery was because XuBP in the incubation mixture was carboxylated to form 3-phosphoglycerate. The concentration of XuBP half-saturating the XuBP-carboxylation reaction of RuBisCO was 12 to 15μM. The initial inhibition and the subsequent recovery of the activity were due to the elimination from and re-binding to RuBisCO, respectively, of the activator CO_2.
