Effects of Ammonia on Respiration, Adenylate Levels, Amino Acid Synthesis and CO_2 Fixation in Cells of Chlorella vulgaris 11h in Darkness

The effects of NH_4Cl on respiration, adenylate and free amino acid levels as well as dark CO_2 fixation were investigated using nitrogen-starved Chlorella vulgaris 11 h cells with or without addition of methionine sulfoximine (MSX), an inhibitor of glutamine synthetase. Upon addition of NH_4Cl (1 mM) to the cells not treated with MSX, respiration was stimulated and the level of ATP dropped rapidly, while the levels of ADP and AMP increased. NH_4Cl also stimulated amino acid synthesis, especially of glutamine, and markedly enhanced dark CO_2 fixation. Addition of NH_4Cl to MSX-treated cells stimulated respiration and lowered the level of ATP, but did not enhance glutamine synthesis and only slightly stimulated dark CO_2 fixation.
