Intact mesophyll protoplasts from cereal plants were easily prepared by incubating leaves with the abaxial epidermis peeled off at 20-25℃ for 2-3 hr in 0.6 M mannitol containing 1% cellulase at pH 5.6. From one gram (fresh weight) of leaves 1.5-6×106 protoplasts, more than 90% of which were morphologically intact, could be obtained. Protoplasts isolated from wheat, oat, corn and barley were efficiently infected with brome mosaic virus (BMV), and supported viral multiplication.
