Intraspecific Variations of Mitochondrial Cytochrome Oxidase I Sequence in an Aphidophagous Species, Aphidoletes aphidimyza (Diptera : Cecidomyiidae)

Aphidoletes aphidimyza, an aphidophagous species of Cecidomyiidae, has been used in many countries as a biological control agent against aphids in greenhouses. The species had been reported previously in Japan. Recently, preliminary tests started in Japan to evaluate its effect on aphids using mass-produced cocoons imported from the Netherlands and United Kingdom. There are no clear diagnostic morphological differences at the subspecies level between Japanese and European specimens. Therefore it is necessary to study intraspecific variations at the DNA level before Japanese populations are contaminated by mating with individuals imported from foreign countries. We sequenced part of the mitochondrial cytochrome oxidase subunit I (COI) gene in twelve Japanese local populations and one commercially imported population from the Netherlands. The sequences were found to be highly variable between Japanese and European specimens.
