The present experiments were undertaken to know the osmoregulative function of the contractile vacuole in the fresh-water Protozoa, Cothurnia sp. Organisms were kept in a continuous flow of the culture medium and then in flow of an experimental solution in the way described by Kitching ('34), Distilled water, Ringer's solutions of various concentrations or those containing alcohol, ether or diluted KCN were used respectively as an experimental solution. During the experiments both the diameter (the maximum diameter just before the systole) and the time interval between two successive systoles of the contractile vacuole were measured. Then, the frequency of the systole and the rate of output from the contractile vacuole were calculated. The following results were obtained. (1) The frequency and, sometimes, the diameter of the contractile vacuole decreased gradually to a new steady value when the organism was placed in hypertonic Ringer's solutions, and the frequency increased in distilled water. (2) The rate of vacuolar output decreased in hypertonic solutions and increased in distilled water. (3) The frequency, diameter and the rate of output returned gradually to their original values when the organism was replaced in the culture medium. (4) When the organism recovered in the culture medium was treated again with hypertonic solutions of the same concentration, the decrements in the frequency, diameter and rate of output became less than those in the former experiment. (5) The cyanide solution inhibited the activity of the contractile vacuole: the frequency and the rate of output decreased to very low values, and, as a result, the body volume increased. Alcohol or ether accerelated the activity of the contractile vacuole and reduced the body volume. From the above results, it seems that the contractile vacuole of Cothurnia sp. acts as a controller of the internal osmotic pressure.
