The present experiments were undertaken to know the effect of Na, K, Mg and Ca ions on the contractile vacuole of the fresh-water protozoa, Cothurnia sp.Experimental method was similar to that descrided by Kitching ('34). Experimental solutions used were 0.01 M Nacl, 0.01 M KCl, 0.006 M MgCl_2, 0.006 M CaCl_2 solutions (pH 6.8-7.0) and mixtures of these solutions. The area at the vertical section of the organism and the diameter of the contractile vacoules (the maximum one just before the systole) and the time interval between two successive systoles were measured in the experimental solutions under the microscope. Then the frequency of systole and the rate of output of the vacuole were calculated. These values were compared respectively with those before the experiments. The following results were obtained. (1) When the organism was placed in solutions of single salt, the frequency of the contractile vecuole decreased, the rate of decrement following the order: K^+<Mg^<++><Na^+<Ca^<++>. (2) The diameter of the vacuole did not vary in NaCl solution but it decreased to a new steady value in othersalt solutions, the rate of decrement following the order: Ca^<++><Mg^+<K^+. (3) the rate of output from the vacuole decreased, the rate of decrement followingthe order: K^+<Na^+<Mg^<++><Ca^<++>. (4) The area at the vertical section of the organism decreased but there were no remarkable differences among the four salt solutions. (5) When the organism was placed in mixtures of these salt solutions, the frequency, diameter and output of the vacuole and the area at the verticalseetion of the organism decreased. The order of ions in the effect on the respective decrement was similar respectively to that obtained by single salt solutions. (6) It is considered that there are antagonism between Na or K and Mg or Ca ions concerning the effect on the frequency of systole and on the rate of outputof the vacuole. (7) It is condidered that the rate of output from the vacuole has some relation to the permeability of the membrane to water and to the gel/solratio of protoplasm.
