The present experiments were undertaken to know effects of cyanide on thecontractile vacuole of the fresh-water protozoa, Cothurnia sp. Experimental method was similar to that described by Kitching ('34). Potassium cyanide and sodium cyanide solutions of various concentrations were used as experimental solutions. During the experiments both diameter (the maximum just before the systole) andtime interval between two successive systoles of the contractile vacuole were measured and the frequency of systole and the rate of output of the vacuole were calculated. The following results were obtained. (1) When the organism was placed in cyanide solutions, the frequency and the rate of output of the contractile vacuole decreased and body valume increased. (2) Decreases in the frequency and in the rate of output were correspondent with the concentration of cyanide solutions. (3) In the same concentration, the rates of decrease in the frequency and in the output of the vacuole which was shown in sodium cyanide were larger than those in potassium cyanide. (4) Diameter of the vacuole increased in the cyanidesolution, and the rate of its increase was larger in sodium cyanide than in potassium cyanide of the same concentration. (5) It is considered that cyanide blocks some respiratory oxidases and so cuts off the supply of energy necessary for systole of the contractile vacuole.
