When mature unfertilized eggs Acheilognathus limbata and A. cyanostigma are added to the sperm artiffcially, spermatoza become very active around the micropyle membrane where a remarkable aggregation of spermatozoa occurs. In some eggs, however, the surface of the vegetal pole also induces sperm aggregation, although not conspicuous as in the case of the micropyle area. Such aggregation on the vegetal pole takes place in the eggs which have come in contact with the animal pole of the egg located just in front of the former, when they are oviposited through the elongated ovipositor making a single row with the vegetal pole of each in front, while any non-contacted egg failed to aggregate spermatozoa on the vegetal pole. The factor responsible for sperm activation and aggregation can be transferred to the intermediate area other than the both poles of the egg membrane, which have come in contact artificially with the micropyle area of another mature unfertilized egg. These facts show that the factor is probably a chemical substance. The factor is not species-specific to heterologous spermatozoa of fish belonging to either different genera or families. But no aggregation of spermatozoa can be observed around the micropyle of eggs of the minnow, the gold fish, the mud loach, etc., all of which spawn both eggs and spermatozoa simultaneously into the fresh water when the male chases the female. The factor located around the micropyle membrane can readily be removed within few minutes by treating the eggs with acidified distilled water, while alkalinized water is not effective. However, spermatozoa aggregate remarkably around the micropyle area when the eggs are exposed to acid side distilled water and inseminated. On the contrary, in alkaline side water the eggs fail to aggregate spermatozoa. These facts may be interpreted as indicating that the factor dissolves into acid water, attracting spermotozoa toward the eggs.
