The present study was performed to investigate the regulative potency of the presumptive lateral mesoderm with regard to the differentiation of the primordial germ cells. Middle gastrulae of Triturus pyrrhogaster and Hynobius nebulosus and early neurulae of Hynobius nebulosus were used. The presumptive intermediate mesoderm was extirpated unilaterally or bilaterally (cf. Table I). Almost of all operated embryos developed normally, however, a few failed to complete gastrulation. Microscopical observation revealed that in most cases, mesonephros and primordial gonad with primordial germ cells differentiate normally in the operated region. In a few cases, the primordial germ cells does not differentiate, and in such cases the defect of mesonephros and the general retardiation of development are simultaneously found. The cause which brought about the defect of the primordial germ cells may be attributed to the disturbance of the normal morphogenetic movement. The present results seem to indicate that the intermediate mesoderm is not determined as prospective primordial germ cells at neurula stage, but may be determined epigenetically in later development as the morphogenesis of tissue environment proceeds.
