Electron-microscopic observations of the developing gonads of 5-, 7-, and 10-day chick embryos were made. Particular attention was paid to the fine structure of cortical cells, germ cells and somatic cells, in the left gonad. The genetic sex of a 5-day embryo was identified by light-microscopic inspection of the right gonad which was taken from the same embryo and cultured on agar medium for 5 days. No remarkable difference in the fine structure of the gonadal cortex between males and females of 5-day embryos could be observed. The germ cells at this stage were characterized by their large size and many pseudopodiumlike projections which were in contact with their surrounding somatic cells. The mitochondria of germ cells were round and usually larger than those of somatic cells. The sexual differentiation of the gonad was recognized in 7-day embryos, where the ovarian cortex was about 30μm in thickness, and the testicular cortex, 15-20μm. In the cytoplasm of germ cells from 7- and 10-day female embryos, elongated mitochondria and many fat granules were observed. Cortical germ cells in the developing testis from 10-day embryos showed some degenerating process.
