Exchange-transplantation of the presumptive ectoderm between X-ray irradiated gastrula of Triturus pyrrhogaster and the normal ones was performed. Dosages of X-rays employed were 50r, 100r and 200r (140kV, 3mA). When the similar ransplantation was done on the non-irradiated embryos, i.e., in the case of control series, seven out of eleven specimens became normal gastrulae and in four cases their gastrulation were disturbed, however in each case further development was continued. In embryos which received irradiated presumptive ectoderm, 64.5% were suppressed in their invagination and 29.1% were abnormal gastrulae and 6.4% were normal; on the contrary, in irradiated embryos whose original presumptive ectoderms were removed and which received untreated ectoderm, percentages of the cases above mentioned were 11.6%, 69.2% and 19.2% respectively. The difference appearing between two experimental series is remarkable. Histological study of the preparation reveals that the defects induced by X-irradiation occurred in the morphological differentiation rather than the histological, and there were found some degenerated cells with pycnotic nuclei and pigment granules mostly in yolk mass. Those cells are considered to be ectodermal cells demaged by X-ray treatment and to sink from blastocoel roof into yolk mass, because such abnormal figures of cells were observed abundantly in the embryos which received irradiated presumptive ectoderm. It may be concluded that the disturbance of gastrulation was not brought about by the restriction of blastoporal function, but rather by the suppression of the morphogenetic movements of the presumptive ectoderm.
