The purpose of this study is to investigate the individuality of chromosomes in the tumor stem cells of the MTK-sarcoma II of the white rat after transformation into the subcutaneous solid form. The subcutaneous transplantation of the tumor ascites from the MTK-sarcoma II derived solid tumors. These solid tumors were injected, after being crushed, in the peritoneal cavity, and this treatment resulted to induce again the ascites tumors. In these ascites tumors, the individuality of the tumor stem cells was investigated in comparison with that of the original ascites tumors of the same strain. The chromosome numbers of the tumor stem cells observed here showed subdiploid 40 in most cells along with 41, 42 ranking next in occurence. In every cell the ceromosome complex is provided with two distinct sets. One of them consists of rod-shaped chromosomes ranging from 28 to 30 in mumber, and the other comprises V- and J-shaped elements of varying sizes, about 10 to 12 in number. In addition to these chromosomes, there occures a prominent V-shaped element of a large size which is always single. Evidence obtained indicates that the chromosomes of the tumor stem cells of the MTK-sarcoma II after transformation into the subcutaneous solid form are fairly identical with those of the original ascites tumors after successive transplantations. BY way of conclusion, it can be said that the individuality of the tumor stem cells has remained unchanged without undergoing any visible transformation through the transplantation experiments undertaken in this study.
