The reproductive organs from 962 females were dissected out in a saline solution under a binocular microscope at varying times from immediately after copulation to about 40 days thereafter. The 2 minute specimens showed the enlarged uterus with a large mass which contained a number of sperms and small granular-like substances. Some sperms were found in the seminal receptacle. Fifteen minutes after copulation, a larger reaction mass was found in the uterus. Some motile sperms were observed in the seminal receptacle and spermathecae. The reaction mass grew to maximum size one hour after mating and disappeared by 48 hours. At that time the uterus was in normal condition. When the reaction mass was placed in saline solution, the sperm became motile. Motile sperm remained alive in the seminal receptacle from 2 minutes to 360 hours, and in spermathecae from 5 minutes to 480 hours after copulation. In saline solution sperms in the storage organs could move. A single mating female was able to produce fertile eggs for about 20 days. The stored sperms progressively decreased and were completely gone within 20 days. The sperms occurring in the seminal receptacle disappeared prior to the disappearance of those in the spermathecae. Drofophila niglomaculata belongs to a type of species showing a strong insemination reaction in homogamic matings.
