Morphological responses of melanophores in isolated scales of Oryzias latipes to normetanephrine, metanephrine, methonamine and mescaline were investigated. Normetanephrine (5×10^<-5>-10^<-3>M) caused a concentration of the pigment granules within both innervated and denervated melanophores. However, the activity of normetanephrine was markedly less than that of noradrenaline or of octopamine. The activity of metanephrine was also much less than that of adrenaline or of sympatol. The sensitivity of melanophores to these drugs was not significantly altered after denervation. Methonamine (10^<-5>-10^<-3>M) induced concentration of the pigment within innervated melanophores, but failed to do so within denervated ones. When methonamine at high concentrations was applied to melanophores, thep igment granules were observed to disperse within the cells. Mescaline (10^<-6>-10^<-3>M) could not induce a concentration of the pigment in the melanophores. This drug, at high concentrations (10^<-4>-10^<-3>M), exhibited an inhibition of the concentrating response of the melanophores which was evoked by K ion. The evidence tends to confirm the conclusions previously submitted that the catechol nucleus in adrenaline molecule has an essential role on a direct action for pigment concentration, and that a substitution of OH group in the phenyl ring causes a transition of effective compounds into those with concentration-inhibitory action.
