The responses of melanophores in the isolated scales of Oryzias latipes to four catecholamines, i.e. noradrenaline (NA), adrenaline (A), isoproterenol (ISO) and dopamine (D), and dichloro-isoproterenol(DCI) were quantitatively examined. All of the compounds except DCI induced concentration of pigments within both innervated and chronically denervated melanophores. Furthermore, the denervated melanophores were found to be supersensitive towards these amines. These results indicate that the catecholamines tested may act directly on the melanophores. The order of relative potency of the above catecholamines for causing pigment concentration and sensitivity change was NA and A> D> ISO on both innervated and denervated melanophores. DCI showed a weak concentrating effect only on innervated melanophores, indicating that it may also act indirectly on the cells. This drug inhibited the concentrating response of the melanophores by K ion, A and ISO. The following conclusions were reached regarding the relationship of chemical structures of these catecholamines to the direct action on melanophores. 1) N-isopropylation in the terminal amino group reduced the direct action. 2) The OH group on the beta carbon atom of the side chain may be important for contributing to the direct action, although it may not necessarily be essential for the action. 3) The phenolic OH groups on the phenyl ring may be essential for the direct action.
