The time course of changes in sensitivity of the scale melanophores in Oryzias latipes to adrenaline, sympatol and tyramine was examined during degeneration and regeneration of their pigment concentrating nerve. The process of functional degeneration as well as that of regeneration of the nerve fibers previously severed was judged from the responses of the melanophores to K ions, which act selectively on the endings of the pigment concentrating nerve fibers to bring about the concentration of the pigment granules. Functional degeneration of the severed nerve fibers took place at the outset of the final dispersion phase and their degenerative state continued for one week. In some scales, the signs of regeneration came 7 days after the operation and within 2 weeks regeneration was completed in almost all the cases. Supersensitivity to adrenaline of the denervated melanophores began to develop from the concentration phase and reached its maximum level at the late stage of the final dispersion phase. The increased sensitivity was maintained with no observable changes during the period of degeneration of the nerve fibers. With regeneration of the nerve supply the increased sensitivity began to decrease and the sensitivity returned to the level in the innervated melanophores when a complete functional innervation of the effectors was attained. With sympatol and tyramine, an increased sensitivity of the melanophores occured transiently on the concentration phase. After the concentration phase, the sensitivity to these amines began to decrease and reached its minimum value at the late stage of the final dispersion phase. The decreased sensitivity continued with no observable change till the functional connection of the melanophores with the nerve reestablished. It was shown that there was a close relationship between the development of denervation supersensitivity to adrenaline and degeneration of the nerve terminals.
