It has been suggested that colloidal iron-positive granules are accumulated toward the surfaces of blastomeres in accordance with the mitotic cycles of Tubifex eggs, and that the granules from vacuoles on the surfaces or at the region of the cleavage furrows (Matsumoto, 1968). In the present observation, various histochemical tests were applied to the material in the vacuoles: Histochemical tests included were staining with sudan Black B, Barrollier's silver reaction, peracetic acid-Schiff reaction, Baker's acid haematein test, stainings with Nile blue, with toluidine blue (pH 4.4), and with Alcian blue, and Muller-Hale's colloidal iron reaction. Colloidal iron reaction was progressively weakened with prolonged fixation with Orth's fluid containing dichromate. A good result was obtained by a fixation for four to seven days. Treatments for a week or more was destructive. It was necessary to avoid any kind of fat solvents for demonstrating the mucosubstance after formaldehyde fixation. In the case of post oxidation with dichromate in the Orth's fixative, the substance became more stable. In formaldehyde-fixed embryos, the mucosubstance was stained strongly with the colloidal iron reaction, but staning with Alcian blue for acid mucopolysaccharide and tests for fats were negative. In dichromate-fixed embryos, staining with sudan Black B, Barrollier's silver reaction, peracetic acid-Schiff reaction, and acid haematein test were also negative. On the contrary, deep blue color was developed by staining with Nile blue, metachro matic dark-brown color by Toluidine blue and bluish green color by Alcian blue. After postchromation of embryos, dark-brown color was observed by Baker's acid haematein test for phospholipid. After rinsing with distilled water for three minutes and staining for one minute, positive colorations with Nile blue, Toluidine blue and Alcian blue were obtained in dichromate-fixed specimens. From the results obtained on the staining ability of the secreted mucosubstance of Tubifex embryo, it concluded that the substance contained both acid mucopolysaccharide and phospholipid.
