In microincineration, general pattern of distribution, size, and arrangement of ash granules depend upon the method of preparation of the specimen. Present paper deals with a comparison of the influence of different fixatives on the ash granules in the spodograms of the eggs of Tubifex hattai Nomura and a cytological observation of the ash distribution in the same material during the stages from cleavage to gastrula. Fixed eggs were sectioned at 7 μ in thickness, incinerated at 520℃ in an electric furnace. Thus incinerated preparations were observed microscopically under the dark-field illumination. In the egg-cells fixed with alcohol or alcholic-formalin mixtures, voluminous and coarse granules of ash of 1 μ or more in diameter were observed. In such cases, the identity of the cellular inclusions was lost in the area of coarse granules. In the cases of Bouin's and Gendre's fluids, some areas of the egg-cell were observed where ash granules were small and homogeneous in distribution. These fixatives also preserved fine structure of the fibrillar elements of the mitotic apparatus. The micromeres, somatoblasts and macromeres in the cleavage stage could be distinguished from each other by the arrangement and deposition of mineral salts. The cytoplasm of the ectoblast 2d and the mesoblast 4d were distinguished clearly from the other blastomeres by an uniform ash distribution and by a small size of the ash particles. The "segmentation cavity" in cleavage stage and the "blastcoel" in blastula stage were destitute of ash particles. The mesoderm in the late gastrula stage was also characterized by a high concentration of ash in comparison with the ectoderm. It was observed in the mitotic apparatus of the micromeres that the spindle area in the metaphase was clearly recognized by its rich deposit of flat white ash, while the area in the anaphase the deposit of ash was poor. The central nuclear sphere contained white ash in a small amount if any. A dense deposit of ash in the equatorial plate of metaphase might indicate that the major part of the combined mineral matter was in the chromatin material.
