In cleaving eggs of aquatic oligochaetes, Tubifex hattai and Limnodrilus willeyi, dark blue spots stained with Muller-Hale's colloidal iron method for acid mucopolysaccharide were observed near the furrow region. The preservation of the mucosubstance could be influenced considerably by different fixing reagents. Alcohol-containing and acidic fixatives had a deleterious effect upon the preservation of the substance. Non-acidic formalin and iodate- or dichromate-containing formalin solution gave satisfactory results. A pH above 6.0 generally seemed to favour the fixation of the substance. An accumulation of colloidal iron-positive granules was observed periodically in parallel with the mitotic cycle of the first and the second cleavages of the egg. Prior to the formation of cleavage furrow the cytoplasm showed a weak blue reaction as a whole, while blue spot on the surface of the egg appeared simultaneously with furrowing of the first cleavage. Further, the formation of the spots began first at the furrow, and spread to the pole region of the egg. After the emergence of the spots the blue color of the cytoplasm disappeared. Then, the number of the spots started decreasing, and when the two blastomeres flattened adhering to each other, two or three of large blue spots were observed near the furrow. The cytoplasm at this time showed a weak blue reaction. In the second cleavage, the same processes were repeated although the blue-colored blastomeres and spotted blastomeres with white-colored cytoplasm were variously in mosaic. In the sectioned specimens of cleaving eggs, colloidal iron-positive granules measuring about 0.5 μ in diameter were accumulating toward egg surfaces, and sometimes densely stained vacuoles were observed inside of the cell membrane. From the present observation it is assumed that the accumulation of the granules in the cytoplasm is followed by the formation of vacuoles at the site of emergence of granules and that the mucosubstance of the vacuoles is secreted into the perivitelline space.
