Scale Melanophores of Zacco temmincki ; A Preparation Suitable for Physiological or Pharmacological Studies on Fish Melanophores

In a cyprinid fish, Zacco temmincki melanophores occur in the dermis surrounding both sides of the scale layer. In an isolated scale, the melanophores located in the dermis under the scale layer are free from the epidermis-covering and are only screened with xanthophores, a layer of iridophores and loose connective tissue. The tissue organization around these melanophores is similar to that around the melanophores in the split fin preparations reported by Fujii (1959), Fujii and Novales (1969) and Fujii and Miyashita (1975). K ions and catecholamines caused melanosome aggregation in the melanophores. Melatonin was very effective in inducing pigment aggregation in the melanophores. The responses to K ions and these drugs proceeded rapidly and simultaneously among the melanophores. The recovery following removal of the stimulation proceeded rapidly and simultaneously. The denervated melanophores were obtained by immersing the isolated scales in a physiological solution over 15 hours (20℃). Based on these results, it was suggested that the isolated scale of this species, especially from the area within the dark band was a preparation suitable in studies on the effects of chemical substances on fish melanophores.
