Distribution of Dendritic Cells in the Lymphoid Tissue of lnterleukin 2 Receptor γ Chain Knockout Mouse

The interleukin-2 receptor y chain is indispensable for IL-2-, IL-4-, IL-7-, IL-9-, and IL-15-mediated signaling. So the lack of this chain induces the severe combined immunodeficiency (SCID). The IL-2 receptor knockout (IL-2R KO) mice showed a decrease in the number of lymhocytes and increase in that of monocytes. B cells were also reduced unlike human XSCID. However it is not clear for the dendritic cells to have been located in KO mice. In this investigation, the distribution of dendritic cells in IL-2R KO mouse was observed by immunohistochemistry. MIDC-8 positive cells were observed in paracortical area of the type 1 KO mouse and in all over the lymph node of type 2 KO mouse. There was hard to separate the cortical area from the medulla in type 2 KO mouse. FDC M2 positive reaction was seen only in the follicle of type 1 KO mouse in which the paracortical area was able to distinguish from the follicle. Germinal center dendritic cells (CD4+CD11c+) were observed in both type KO mouse. In normal mouse, FDC and GCDC are located in germinal center, but these two dendritic cells showed different location in the KO mouse lymph node. This suggests that these cells may have a different origin and life span.
